From time to time, i do use the remote on the wall and that can mess up with the ESPSomify.

Is there a way to not disable buttons if the status in HA is 100 opened/closed? I want to be able to press up/down any time and to get it moving despite the status.

If it is oppose the status, it can just reset the position at that moment.

Unfortunately, I am not the one disabling those buttons. It is baked into the cover component in HA.

However, it sounds like you did not link the wall remote to the shade defined in ESPSomfy RTS. If you do that it will not matter how the motor was moved.

I have two physical remotes for the shades, the espsomify is the third one.

Link them in ESPSomfy RTS and it will hear any remote (up to 7 of them) to keep the position correct. If the motor is part of a group don't forget to link that as well.
